抑制GbMIOX5基因表达降低海岛棉对大丽轮枝菌的抗性

摘 要:

由大丽轮枝菌(Verticillium dahliae)引起的黄萎病病害是影响海岛棉的生长发育的重要因素, 对棉花产量和品质等造成严重影响。虽然前人研究已经鉴定了一些参与棉花抗病的基因和蛋白, 但是抗病的机制仍不清楚。本研究从海岛棉中鉴定获得一个编码肌醇加氧酶(myo-inositol oxygenase)的基因GbMIOX5表达分析表明GbMIOX5显著受到大丽轮枝菌的诱导表达。为了分析GbMIOX5参与病原菌响应的功能以海岛棉CLA1 (Cloroplastos alterados 1)为标记基因, 利用烟草脆裂病毒(tobacco rattle virus, TRV)诱导的基因沉默技术构建沉默载体sTRV:GbMIOX5并转化海岛棉。 GbMIOX5沉默植株中GbMIOX5的表达水平与对照相比显著降低; 同时GbMIOX5沉默植株对大丽轮枝菌的抗性显著降低, 并表现出H2O2的显著累积及D-葡萄糖醛酸和抗坏血酸含量的显著降低。

Abstract:

Verticillium wilt generated by Verticillium dahliae is an important factor to affect the growth and development of Gossypium barbadense and leads to great reduction of fiber yield and quality. Although many studies of genes and proteins involved in the interaction between cotton plants and V. dahliae have been reported, however, the molecular mechanism of disease resistance is still unclear. In this study, a myo-inositol oxygenase (MIOX) gene GbMIOX5 was identified in G. barbadense. Expression analysis showed that GbMIOX5 was significantly induced by V. dahliae. In order to analyze the function of GbMIOX5 in response to V. dahliae, the vector sTRV:GbMIOX5 was constructed using tobacco rattle virus (TRV) mediated virus induced gene silencing (VIGS) technology, and was then transformed into G. barbadense. The expression of GbMIOX5 was significantly suppressed in VIGS plants of G. barbadense. Compared with the control cotton plants, the VIGS plants of G. barbadense indicated significant resistance reduction to V. dahliae, coupled with increase of H2O2 content and decrease of D-glucuronic acid (D-GlcA) and ascorbic acid (AsA) content.
